1/ Nomenclature des anticoprs de synthèse :

1975 - momab – 100% Murin

ex : Anti-CD3 muromomab/OrthocloneOKT3® est un médicament immunosuppresseur administré afin de réduire le rejet aigu chez les patients ayant eu une greffe d'organe.

Il a été le premier anticorps monoclonal à être approuvé pour une utilisation clinique chez l'homme.

1984 – ximab– anticorps chimère : beaucoup moins immunogène

ex : Rituximab/Mabthera®, Cétuximab/Erbitux®, Infliximab/Remicade® (61% de HAMA pour Infliximab)

1989 – zumab – anticorps humanisés

ex : Daclizumab/Zenapax® (34% HAHA), Alemtuzumab/MabCampath® (1.9% HAHA) or Transtuzumab/Herceptin® (>0.1% HAHA)

1999 – mumab– anticorps complètement humain

ex : Panitumumab/Vectibix®

HAMA : réaction immunogène d'anticorps humains antimurins

HAHA : réaction immunogène d'anticorps humains antihumains

2/ Anticorps conjugués :

anticorps conjugués à une seconde molécule : une toxine, un radioisotope radioactif ou un tag (ex : fluorophore)

exemples de radiomarqués :

Tositumomab marqué par Iodin(131) / Bexxar®,

Ibritumomab tiuxetan/Zevalin® marqué par Yttrium(90) ou Indium(111),

exemple de toxine conjuguée :

Gemtuzumab ozogamicin / Mylotarg®
